酸素結合蛋白質の分子操作と精密構造機能研究の新展開

研究概要	種々の酸素結合蛋白質の機能発現分子機構の研究の新展開をめざし、以下の結果を得た。1.ヘム近傍のアミノ酸残基の人工変異によるヘモグロビン(Hb)の酸素親和性の制御(今井)、2.対称型Fe-Zn混成Hbの酸素平衡測定による微視的酸素平衡定数K_1,K_2値の決定(森本)、3.分子内ジスルフォイド結合を導入したミオグロビン(Mb)のリガンド結合速度およびヘムポケット内のLeu-29の機能的役割の解明(森島)、4.組換え技術による根粒菌の酸素センサー蛋白FixLの第五軸配位子の同定など、ヘムの配位構造の解析(飯塚)、5.紫外共鳴ラマン分光によるHbNO(+IHP)のT状態とdeoxyHbのT状態の間のα1-β2接触面での構造の違いの検出(北川)、6.天然・人工変異体、化学修飾体を用いた、T状態マーカーである287nmでの負のCDバンドに寄与するアミノ酸残基の同定(長井)、7.Mb結晶のX線回折強度データに基準振動精密化法を適用して得られた蛋白質の揺らぎの温度依存性と中性子散乱の結果との比較(郷)、8.種々の光学的プローブを用いたストップドフロー法による、Mbのフォールディング過程でのヘムの役割(木原)、9.ポルフィリン環のメソ位窒素原子導入はMbのリガンド結合機能に劇的な影響を与えること(根矢)、10.環形動物多毛類のHbの階層的構造を安定化するリンカー鎖の分子内の位置の決定(後藤)、11.インドールアミン2酸素添加酵素に相同のサザエのMbの自動酸化性、吸収スペクトル測定とそれらの分子進化的意義(鈴木)、12.フトヘナタガイのMbの自動酸化は二量体→単量体変化を伴うこと(四釜)、13.胎児ヘモグロビンは低酸素環境下で効率の良い酸素運搬を、成人ヘモグロビンはボ-ア効果が最大限に発揮できるように、それぞれ最適化されていること(小林)。
