研究概要

スキンドファイバーを用いる、筋線維の力学特性の研究は、ケージドヌクレオチドおよびケージドカルシウムなどの開発によって、可能となった。山田は筋線維に蛍光標識を行い、ケージドATPからのATPの急速遊離によって生じる蛍光変化の時間経過から、ミオシンのアクチンとの相互作用の存在とその特性を明かにした。杉はこの方法によって、化学・力学エネルギー変換効率の荷重依存性の研究を行い、個々のミオシン頭部が収縮の初期に荷重を感知して、エネルギー変換効率を決定していることを示す結果を得た。平塚は筋線維を対象とする研究に有用な蛍光ATPアナログを開発した。荒田はミオシン頭部の根元の屈曲によって収縮力が発生するとの仮説を検証する為、この部分にスピンラベルを行ったが、収縮時のラベルはランダム配向を示し上の仮説と対立する結果を得た。このように、ミオシン頭部の首振り運動仮説が立証されないことから、堀田はアクチンの変異した筋線維を多種類得ることを目的とし、ショウジョウバエ突然変異17系統の中から、いくつかの生理学的に重要な系統を提示した。片山は滑り運動を起こしているミオシン・クロスブリッジの一瞬の姿を捉えることを目指して、高い空間、時間分解能をもつマイカ細片・急速凍結電顕法により、硬直、弛緩、収縮クロスブリッジを観測した。石渡は光ピンセットによるアクチン・ミオシン間の相互作用の一分子力学・熱力学研究を行った。若林、八木、馬詰は、ミオシン頭部とアクチンの相互作用によって生じる構造変化のX線回折による研究を行った。アクチンフィラメントの弾性的伸展と同時にアクチンの構造変化が検出された。柳田は、ミオシンの一つの分子がATPを分解し力を発生する過程を直視下に捉えた。飯野は興奮収縮連関機構について、リアノジン受容体には骨格筋型および親近型などのサブタイプがあり、それぞれ機能に差異のあることを示した。呉林は筋線維中のリアノジン受容体について、カルシウムによる制御の有無を調べた。大槻は収縮のカルシウム調節機構について、とくにトロポニンT分子のC端部分がその働きを担っていることを示した。太和田と児玉は筋収縮の理論的および熱力学的解析を進めた。
