研究概要

平成8年度は以下の成果が得られた。
(1)新しいNCaP遺伝子cDNAをクローニングできた。それらはカエル嗅上皮p26olf、錐体視細胞中のs26、ラット海馬に存在するHLP3とHLP4、分裂酵母第一染色体の存在する機能未知の蛋白質などである。
(2)ヒポカルシンについてゲノム遺伝子を単離するとともにヒトの第2染色体に存在することを明らかにした。
(3)S-モジュリンの標的蛋白質がロドプシンキナーゼであることが同定された。ニューロカルシンの標的蛋白質候補として、S100bとチューブリンb鎖が考えられた。
(4)脂質によるリカバリンの修飾の生理的役割について、ロドプシンキナーゼを細胞膜によりよくアンカーする作用があることが明らかになった。また、S-モジュリンとs26において見られる脂質膜への結合性の違いはC末端のアミノ酸の違いに原因があることが明らかになった。
(5)ムスカリン性アセチルコリン受容体に対するGRK2によるリン酸化がカルモジュリンまたはS100蛋白質によって調節されることが分かった。
(6)キメラカルモジュリンを作成し、それぞれのドメインがどの様な作用を持つのか調べた。その結果、一対のカルシウム結合サイトが協同的に機能するときのみ、活性を持つことが分かった。
(7)カルシニューリンの役割として、記憶のモデルである長期増強現象を抑制的に制御している可能性があることが明らかになった。また、分裂病モデルラットを使った実験から、NVPの分裂病の病態における役割を示唆する結果を得た。
