研究概要	(1)経口免疫寛容の利用各種T細胞抗原決定基ペプチドを合成し、その経口免疫寛容誘導能を検討した。その結果、ウシα_<s1>-カゼイン91-110は抗原特異的T細胞増殖応答を抑制し、β-ラクトグロブリン119-133については抗体産生応答の抑制が認められた。II型コラーゲン245-270はコラーゲン誘導性関節炎の発症を部分的に抑制した。抗原の投与量についてはそれぞれ至適値があった。前2者についてはタンパク質分解酵素により分解されやすくII型コラーゲン245-270については比較的消化耐性であった。 (2)抗原分子のアミノ酸置換アナログの利用ウシβ-ラクトグロブリンの優勢なT細胞抗原決定基を含むペプチド119-133(p119-133)の各アミノ酸残基を置換したアナログペプチド(18種)を化学合成し、MHCクラスII分子との結合親和性を測定した。その結果、126残基目のPro残基と128残基目のVal残基がMHCクラスII分子との結合に重要であることが明らかとなった。p119-133に特異的な4種のT細胞クローンを樹立し、それぞれのアナログペプチドに対する免疫応答を調べた結果、122-129領域のアミノ酸残基のうち上記の2残基以外は全てT細胞レセプターとの結合に関与することを明らかにした。T細胞クローンのp119-133に対する応答を抑制する活性をもつアナログペプチドを検索したところ、129残基目のAspをAlaに置換したペプチドD129Aは4つのクローンのうち3つに対して抑制活性をもつことが明らかとなった。このペプチドをp119-133と同時にマウスに非経口的に投与したところ、p119-133に対するT細胞応答、抗体産生応答ともに抑制された。
