研究概要

単細胞緑藻(Scenedesmus acutus)を用いた実験で確認された,“プロトポルフィリノーゲン-IX酸化酵素の阻害,ポロトポルフィリン-IXの蓄積,エタン発生をともなうチラコイド膜の破壊,光合成色素の減少"といういわゆるPeroxidizing植物毒性作用の解明を目的として,科学研究費交付最終年度の検討を行い,以下の成果〈[1]〜[4]〉を得た.
[1]HPLC-イソルミノール化学発光を原理とする全自動脂質分析システムを用い,プロトポルフィリノーゲン-IX酸化酵素阻害剤処理後に生ずる過酸化脂質を測定し,活性酸素が関与するチラコイド膜破壊の機構を明らかにした.
[2]クロロフィル生合成能を有する植物培養細胞を用いた生物試験を行い,上記のPeroxidizing植物毒性作用が緑色植物の葉緑体中で普遍的に起こることを確認した.
[3]光存在下で活性酸素を発生させチラコイド膜破壊を誘導する新しいプロルフィリノーゲン酸化酵素阻害剤の分子設計と合成を行なった.
[4]分子設計し合成した阻害剤の多くは,各種植物のプロトポルフィリノーゲン酸化酵素を阻害することが確認された.
平成9年度の研究で得られた結果は,(11)項記載の雑誌等に発表した.
