研究概要	本研究では生体防御系における細胞応答とその応答を制御する遺伝子間相互作用の分子機構の解明を目的としている。本年度はまず、我々がインターフェロン(IFN)系の転写制御因子として発見したIRF-1の標的遺伝子群の解析を行い、Interleukin-1β converting enzyme(Ice)を同定した。実際Ice遺伝子は活性化大リンパ球において発現誘導されるがIRF-1欠損マウス由来Tリンパ球では全く誘導が見られなかった。Iceプロモーター領域にIRF-1結合部位を見いだした。更にIRF-1欠損Tリンパ球ではDNA障害によるアポトーシスの誘導が抑制されることを明らかにした。更に新しい標的遺伝子を何種類か同定しつつあり、これらの遺伝子の細胞応答への関与を解明しつつある。一方、IRF-1に加えてIFN系を制御すると考えられる転写因子p48(ISGF3_γ)の欠損マウスの作製に成功した。このマウス由来細胞の解析によって、p48がIFN-α/βの応答のみならずIFN-_γの応答にも必須の役割を果たしていることを明らかにした。p48欠損マウスでは現在のところリンパ球の分化異常が見いだしていない。リンパ球増殖の調節機構を解析するためにインターロイキン-2(IL-2)受容体によるシグナル伝達機構の解析を進め、本年度はbcl-2遺伝子の誘導がJakキナーゼや他の既知の非レセプター型チロシンキナーゼとは独立した経路によって担われていることを明らかにした。
