冠動脈の緊張亢進や内膜肥厚の防止に関する細胞分子生物学的研究

研究概要

本研究は、冠動脈平滑筋細胞の緊張・増殖の制御とその破綻の機序、およびその防止方法を細胞分子生物学的に明らかにすることを目的とする。細胞情報の光学的測定装置とRTPCR法を用いて以下の研究成果を得た。
(1)細胞内小胞輸送阻害物質ブレフェルジンは、in situ 血管内皮細胞において、Caシグナルには影響せずに、ブラジキニンによるEDHF生成/分泌を選択的に抑制した。
(2)細胞間接触が引き起こす培養内皮細胞の増殖停止はストレスファイバーの形態変化(細胞骨格の再構築)を伴うことが明らかとなった。
(3)内皮付豚冠動脈において、トロンビンが引起こす内皮依存性弛緩反応の第1相にはNOとEDHFが、第2相にはEDHFとPGI2が関与すること、第1相とNO産生は良い相関を示すこと、トロンビンによる内皮細胞の一過性[Ca]i上昇は、NO産生と相関することが明らかとなった。
(4)初代培養平滑筋細胞においては、増殖細胞の密度が高くなるにつれ、ATIIに対して[Ca]i上昇反応を起こす細胞の数は増えたが、上昇程度は均一でむしろ低くなった。
(5)初代培養平滑筋細胞では平滑筋型トロポミオシンαの発現が優勢であるが、血清添加により増殖を刺激すると、非平滑筋型トロポミオシンaの発現が増加する。非平滑筋型トロポミオシンaの発現調節にPKCの関与が示唆された。
(6)位置指定変異導入法を用いてカルモヂュリンの4つのCa結合部位のグルタミン酸をグルタミンに置換した変異体を作製し、その機能をスキンド家兎腸間膜動脈の収縮を指標として検討した。血管平滑筋の収縮活性化機構においてはCa結合の第4ドメインがもっとも重要な役割を担っていることが明らかとなった。
