研究概要

表皮ケラチノサイトは細胞外からのシグナルに対応して細胞骨格-細胞接着系の構造分子の集合と離脱を動的に繰り返し、細胞の分化(角化)と増殖を制御している。本研究は自己免疫水疱症、先天性表皮水疱症、角化異常症の発症機序の分子医学的解明をモデルとしてケライチノサイトの細胞骨格-細胞接着系分子構造の機能的制御機構を分子医学並びにシグナル伝達の観点からその病的状態と共に相補的に解明しようとするものである。
昨年度までに天疱瘡IgGが培養ケライチノサイト表面抗原(デスモグリエンIII)に結合することによって、細胞内Ca^<2+>とIP_3の一過性の上昇が見られ(J Invest Dermatol 104:33-37,1995)、この上流にはホスホリパーゼC(PLC)の活性化が関与し、最終的にはプラスミノーゲンアクチベータ-の分泌誘導がある(J Invest Dermatol 105:329-333,1995)こと、さらに、この系においてCキナーゼ(PKC)の活性化、PKCの各種アイゾザイムの細胞膜への特徴的な転移パターンを生じること(J Invest Dermatol 108:482-487,1997)などを示した。今年度は、PKCの活性化に直接関与するダイアシルグリセロールがPLDではなくPC-PLC依存性に産生されることを示した(J Invest Dermatol 109:650-655,1997)。また、天疱瘡抗体依存性燐酸化の基質について、デスモソーム構成分子であるデスモグレイン、プラコグロビンの燐酸化の増強とその分子間結合の離開を発見した。さらにその結果、デスモグレインIIIの崩壊が生じることを見いだした(国際研究学会1998年5月発表予定)。類天疱瘡についてはヘミデスモソームの構成分子であるBP180がホルボルエステル(TPA)で燐酸化され、これに伴ってBP230が細胞質から細胞膜へ転移すること、これはPKC阻害剤(H7)で阻害され、同時にヘミデスモソームから分散することから、ヘミデスモソームの制御にはPKCが関与することを示した。(J Invest Dermatol,revised submitted)。
