低線量・低線量率放射線のヒトへのリスク決定要因に関する基礎的研究

研究概要	低線量・低線量率放射線の身体的、遺伝的リスクを求めるため、ヒトに見られるあらゆる疾病を対象とし、個体および各臓器の線量率効果係数(DDREF)について4年計画で実験を行い、放射線のヒトへのリスク推定の基礎資料とする。 1.低線量・低線量率放射線の身体的リスクとDDREF(Dose and Dose Rate Effectiveness Factor): 放射線感受性および発がん臓器スペクトラムが異なり、ヒトに見られるほぼ全ての種類のがんが検出できる2系統のマウスC57BL/6J、C3H/HeJを用い、1.7Gyの^<60>Coガンマ線を6週齢より7日間隔で計4回(計6.8Gy)を、線量率を変えて(0.57,0.05,0.01,0.003,0.001,0.0002Gy/min)で各100匹に照射した。0.5Gy4回(計2.0Gy)照射についても平成7年度より開始した。24ケ月間の飼育観察を行い、平成9年度に全てのマウスを屠殺する。造血障害および発生腫瘍に関する病理診断を行っているが、1.7Gy4回高線量率照射により高率にリンパ性白血病が発生している。 2.低線量・低線量率放射線の遺伝子リスクとDDREF: 雄N5マウスに^<60>Coガンマ線5.04および2.16Gyを線量率0.57,0.0002Gy/minで照射た。3ヶ月後(精原細胞期照射)に正常雌N5マウスと交配し、子孫(F_1)における致死(流死産)、奇形、機能異常(仮死など)、がん発生を調べる。F_1マウスは1年後に屠殺する。また、ショウジョウバエ翅毛突然変異検出系およびマウス高感度突然変異検出系PT-HTF_1を用い、低線量の^<252>Cf中性子、^<60>Coガンマ線、X線(0.05〜1.2Gy)による突然変異の検出を開始した。 3.遺伝的、身体的リスクのDDREFに関する分子レベルの変化: 上記実験で得られたマウス個体の各臓器、組織、細胞は全て凍結保存し、放射線感受性を決定する遺伝子の変異を迅速に同定するために、専用のDNAシーケンサーを設置し、解析を開始した。
