低線量・低線量率放射線のヒトへのリスク決定要因に関する基礎的研究

研究概要

低線量・低線量率放射線の身体的、遺伝的リスクを求めるため、ヒトに見られるあらゆる疾病を対象とし、個体および各臓器の線量率効果係数(DDREF:Dose Dose and Rate Effectiveness Factor)について4年計画で実験を行い、放射線のヒトへのリスク推定の基礎資料とする。3年度に得られた成果は以下の通りである。
1.低線量・低線量率放射線の身体的リスクとDDREF:放射線感受性および発がん臓器スペクトラムが異なり、ヒトに見られるほぼ全ての種類のがんが検出できる2系統のマウスC57BL/6J、C3H/HeJを用い、1.7Gyの^<60>Coガンマ線を6週齢より7日間隔で計4回(計6.8Gy)を、線量率を変えて(0.57,0.05,0.01,0.003,0.001,0.0002Gy/分)で各100匹に照射し、24ケ月間の飼育観察を終わった。高線量率照射(0.57Gy/分)でリンパ性白血病がC57BL/6Jマウスに62.7%,C3H/HeJマウスに40.4%発生した。線量率を下げるに従い、発生率は低下し、0.0002Gy/分では、C3H/HeJマウスで40分の1、C57BL/6Jマウスで20分の1に低下した(DDREFは約20〜40)。各臓器に固型腫瘍が高率に発生した。高線量率照射(0.57Gy/min)でC3H/HeJマウスで83.2%、C57BL/6Jマウスで20.8%であった。しかし、線量率を下げても、あまり腫瘍発生率は低下せず、0.0002Gy/min照射群で約1.5分の1に減少したにすぎなかった。即ち、DDREFは約1.5となる。
2.低線量・低線量率放射線の遺伝的リスクとDDREF:マウス高感度突然変異検出系PT-HTF_1を用い、低線量の^<252>Cf中性子、^<60>Coガンマ線、X線(0.05〜1.2Gy)による突然変異の検出を行った。現在のところ、突然変異発生率は^<252>Cfで非常に高く、X線と^<60>Coガンマ線には差は認められなかった。中性子線のRBEは、6位と推定している。
3.分子レベルの変化:ヒトにおいてもWT1遺伝子を用いたヒト白血病超高感度検出系を確立し、ヒト白血病の早期発見に有用であることを明らかにした。
