振動分光学による生体NO作用機作の解明

研究概要	一酸化窒素(NO)が生理活性物質として生体中で働いているという報告は、分子科学者にとって全く意外であった。NOは不対電子を持つために反応性が高く、したがってその寿命は短い。NOはアルギニンからヘム酵素により合成され、別の細胞のグアニレートシクラーゼのヘム鉄に結合し、GTPからcGMPを生成する反応を促進することが最近明らかにされた。その機能するNOがNO^+カチオンか、中性ラジカルか、或いはNO^-アニオンかは決まっていないうえ、これが何に代謝されていくかも不明である。本研究では、非常に低濃度にしか存在しない生体NOを赤外分光で検出するために、超高感度赤外分光システムを製作した。これはダイオードレーザーを光源とし、回折格子のような分光器具をつけないシステムであるため非常に明るい。したがって水の吸収に妨害される領域でも、マイクロ秒の時間分解能で赤外吸収スペクトルを与えることが試験測定の結果明らかになった。NO^+、NO、NO^-、NO_2^-などは異なるN-O伸縮振動をもつので、その振動数の観測によりNOの代謝生成物をも追跡する予定である。合成されたNOのレセプターであるグアニレートシクラーゼをウシ肺より単離、精製する実験を開始し、現在本酵素特有の吸収スペクトルを持つ成分が未精製ではあるが得られている。この精製のためには、GTPを結合させたアガロースカラムを自分で用意してアフィニティクロマトグラフをかけることが本質的であることがわかってきた。次年度に継続されることになっているが、第一年目の進展は期待通りで、次の発展が楽しみになりつつある。
