研究概要

ラン藻と葉緑体のRNA結合蛋白質の機能に関する研究を行った。本研究で得られた主な成果は次の通りである。
1 単細胞性ラン藻Synechococcus PCC6301株のRNA結合蛋白質Rbp1とRbp2は細胞質中に局在しRNA分子と結合していることを明らかにした。Rbp2遺伝子をノックアウトしても細胞増殖には影響しないが、Rbp1遺伝子をノックアウトした形質転換ラン藻が得られなかったことから、Rbp1遺伝子がラン藻にとって必須の遺伝子であることが判明した。現在、Rbp1とRbp2のターゲットとなっているRNA成分の分離同定を行っている。
2 葉緑体RNA結合蛋白質の機能を明らかにするため、葉緑体mRNAのプロセシング反応に及ぼす効果をin vitro反応系で調べた。その結果、葉緑体のin vitro反応系からRNA結合蛋白質を完全に除くと、mRNAのプロセシング反応が若干促進することが判明した。RNA結合蛋白質がmRNAのプロセシング反応に対して負の制御をしていることが示唆された。
3 葉緑体RNA結合蛋白質に似た構造をもつ核内RNA結合蛋白質(RZ-1とRGP-3)をコードするcDNAをタバコ属の一種Nicotiana sylvestrisから単離しその推定アミノ酸配列を決定した。RZ-1は核内mRNAに結合して60Sの沈降係数をもつ大きなリボヌクレオプロテイン粒子を形成していることから、前駆体mRNAのプロセシングに関与していると考えられる。RGP-3は10Sよりも軽い粒子として核内に存在していた。今後、植物の核内リボヌクレオプロテイン粒子の構成成分を明らかにするとともに、本研究で示唆されたRGP蛋白質の機能をin vitroで検証する必要がある。
