植物のアルミニウムストレスに対する応答と耐性誘導に関する初期反応の解析

研究概要	A1ストレスは根の伸長阻害として発現する。A1の集積部位を調べると、エンドウ根の場合は根端ほど多く、根幹にむかうと急激に減少した。同様な結果が耐性を異にするコムギでも認められた。A1耐性種(Atlas 66)に比べ感受性種(Scout 66)は、一定濃度のA1で処理すると根端に蓄積するA1は明らかに多かった。これらの結果は根の伸長阻害が根端に集積するA1によることを示唆している。コムギ根の伸長阻害を細胞レベルで調べると、根の伸長域の細胞が特異的に障害を受けていた。すなわち、表皮より2〜3層内側の細胞の縦方向の伸長が著しく抑制され、逆に横方向に肥大していた。根の伸長抑制はA1処理後、3時間目で観察された。原形質膜が破壊(細胞死を意味する)した場合に細胞内に取り込まれ、DNAと結合して蛍光を発する試薬(propidium iodide)を用いると、伸長域の細胞はA1処理後、極めて短時間のうちに死に至っていると考えられた。これら細胞死の直接的な原因は不明であるが、細胞内のpHの低下が有力な理由の一つと考えられた。一方、A1によって引き起こされる細胞死に遅れてリグニンの生成が認められた。また細胞伸長の阻害過程で配向性を持った微小管が消滅しているのが観察された。コムギ、エンドウのA1による根の伸長阻害と細胞死はCa^<2+>によって著しく軽減された。タバコ培養細胞のA1毒性の発現に鉄イオン(Fe^<2+>)が相乗的に関わっていることを明らかにしたが、その機構について調べた。AlおよびFe^<2+>イオンが単独で存在する場合に比べ、両者が共存すると脂質過酸化が約20倍に増加した。一方、種々の抗酸化剤とともにA1とFe^<2+>を共存させると脂質過酸化が抑制され、かつ細胞増殖の阻害も起こらなかった。
