研究概要	酵母が高塩条件(最高1.5M位)に適応して増殖する機構の解析を行なっている。すでにわれわれはCa2^+により活性化されるプロテインホスファターゼカルシニューリンは、通常の生育には必須ではないが、高濃度の塩を含む培地における生育には必要なことを明らかにした。CNは細胞内のNa^+を細胞外に排出し、細胞内のNa^+濃度を低下させるのに必要であった。今年度は、CNの作用機構の解析を行なった。CNは、Na^+排出ポンプをコードする遺伝子PMR2/ENA1の転写を活性化するのに必要である。一方、塩耐性にや細胞内のcAMPレベルが低いことが必要なことを見い出した。cAMP依存性プロテインキナーゼはPMR2/ENA1の転写を負に制御していることが明らかになった。すなわち、cAMPシグナルとCa2^+シグナルは拮抗的に耐塩適応を制御していることがノーザン解析により明らかになった。cAMP依存性プロテインキナーゼとCNは共通の基質のリン酸化・脱リン酸化を通じて転写因子のリン酸化レベルを拮抗的に調節していると思われる。以上の結果は、通常の生育を行なっている酵母が高温ストレスに出会うと、酵母は細胞内cAMPを低下させ増殖を停止すると共に、Ca2^+濃度を一時的に上昇させてNa^+排出ポンプの遺伝子の転写を活性化させることにより高塩条件への適応を制御していることを示している。
