有毒・有害微細藻の遺伝子マーカーによるバイオセンサーの開発と利用

研究概要

本研究では、有毒・有害微細藻類がもつ独特の遺伝子情報を検索することにより、より有効なバイオセンサーを開発し利用しようとするものである。また、微細藻類には特異的なモノクロナル抗体を用いることにより、有毒・有害微細藻類が発生する際のバイオセンサーとしての利用性をもさぐることを目的とする。
1996年度において得られた研究実績は以下の通りである。
○Alexandrium属8種およびその他の渦鞭毛藻、ラフィド藻、緑藻、ユ-グレナ藻、珪藻、ハプト藻、
○Alexandrium属4種22株の5.8SrDNAおよびスペーサー(ITS1、2)領域のPCR増幅産物をすべてクローニングした後に塩基配列を決定したところ、日本産とアメリカ産A.tamarense株8株とアメリカ産A.fundyense株は、その増幅産物に異なる2種のDNA断片をほぼ1:1の割合で保持していることが明らかになった。
○得られた各株の塩基配列を比較した結果、ITS1の3′末端150から170の塩基配列が各ITSタイプ内では相同性が高く、各タイプ間では低いことを明らかにした。
○A.tamarenseとA.catenallaのITS1の3′末端領域おアンチセンス鎖にそれぞれ相補的なPCRプライマーTF1またはCF1とITSプライマーを用いて本属6種の培養株より精製した全DNAを鋳型としてPCRをおこなったところA.tamarenseとA.catenellaの種特異的な増幅シグナルを得ることに成功し、これらプライマーを用いることにより両種を簡便に識別することを可能にした。
○Gymnodinium mikimotoiの細胞からPCR法によって得た18SrDNA領域の塩基配列を他の渦鞭毛藻のそれと比較した結果、Prorocentrum micansと97%、Symbiodinium属各種と約94%の相似性が認められた。
○本研究室で開発したモノクロナル抗体M8751-1を用いてそのA.tamarenseとA.catenellaの各株への反応性を調べたとこと種間の識別がフローサイトメーターを用いることにより可能であることを明らかにした。また、天然のA.tamarense細胞をふくむ海水を免疫細胞分画手法を用いることにより認識できることを明らかにした。
