研究概要

研究計画作成当初の目的は、次の4項目にまとめられる。【1】下垂体前葉でのアネキシンVのホルモンとの共存関係を明らかにすると共に、ホルモン分泌及びホルモン遺伝子の発現調節に対する作用を明らかにする。【2】横体に存在するアネキシンVの作用を明らかにする。【3】視床下部神経活動に対するアネキシンVの影響を検討する。【4】アネキシンVの測定系を開発する。まず、下垂体前葉の隣接切片を作製し、その向かい合う面(ミラーイメージ)を、それぞれアネキシンVとホルモンに対する抗血清で染色するという方法を用いて、下垂体内にアネキシンVの分布を明らかにすることが出来た。この成果については、1996年にサンフランシスコで開催される国際内分泌学会で発表する予定である。更に、下垂体のプロラクチン分泌が、アネキシンVmRNAに対するアンチセンスオリゴヌクレオチドの負荷によって、著しく促進されることを下垂体前葉細胞の初代培養系で明らかにした。黄体では、黄体の機能状態に従って、アネキシンVの分布に変化の見られること、黄体細胞の培養系でアネキシンVがプロジェステロン分泌を促進することを見出している。視床下部スライス培養実験では、アネキシンVの抗体を添加することによって、自発発火の増加する神経細胞の存在することが予備実験で発見されている。アネキシンVの測定法の開発では、現有の抗血清がこの測定法に不向きであることが明らかになり、現在新な抗血清のロットを制作中である。しかし、プロラクチンとLHの測定系は実用段階に達し、下垂体初代培養細胞の放出ホルモンの測定に応用中である。平成7年度に得られた研究結果の概要は以上の如くであり、アネキシンVが視床下部-下垂体-卵巣系で、情報伝達と性腺機能発現に対して重要な役割を担っている可能性の極めて高いことが示された。
