平滑筋細胞の分化機構に関する分子・細胞生物学的解析

研究概要	本研究は培養平滑筋細胞の分化型質維持システムを確立し、平滑筋細胞の形質転換の分化機構を解明することを目的として遂行した。昨年度の研究でラミニン上、インスリン様増殖因子(IGFI、IGFII)またはインスリン存在下では分化型形質維持した状態で平滑筋細胞を培養できることを見いだした。本年度はこの分化型培養平滑筋細胞を用いて平滑筋細胞の分化を誘導する細胞膜を介したシグナル伝達系及び遺伝子発現機構を解析した。分化型形質維持能を有する増殖因子の濃度依存性を調べた結果、IGFIが最も低濃度で分化型形質維持が可能であったことから、IGFIレセプターを介したシグナル伝達系の関与が示唆された。さらに、中和活性を持つ抗IGFI抗体でラミニン/IGFIの持つ分化維持効果及びラミニン単独による脱分化遅延効果が完全に抑制されたことから、IGFIが平滑筋細胞の分化形質維持に関わる因子であることが判明した。このラミニン/IGFI及びラミニン単独の効果はチロシンリン酸化阻害剤またはPI3キナーゼ阻害剤により抑制された。以上の結果からラミニン及びIGFI受容体を介したシグナル伝達系が平滑筋細胞の分化を促し、チロシンリン酸化及びPI3キナーゼがこのシグナル伝達系に関与していることが示唆された。また、この研究過程で、ラミニン受容体であるalb1インテグリンのうちa1インテグリンの発現は転写レベルで制御を受け、平滑筋細胞の分化に伴い増加し、脱分化過程で抑制を受けることを見いだした。さらに、α1インテグリンのプロモーター解析を行った結果、カルデスモン・プロモーターの場合と同様にCArGボックス様の配列が分化型平滑筋細胞での転写促進に必要で、SRF(serum response factor)がコア因子として結合することを明らかにした。また、同じ平滑筋細胞でありながら内臓平滑筋細胞では形質転換に際して、平滑筋型a-アクチンの発現は分化型細胞では低く、逆に脱分化に伴い著しく増加するという血管平滑筋細胞とは逆のパターンを示すが、平滑筋型a-アクチン・プロモーター解析の結果、正の因子として働く2つのCArGボックス様の配列とpurine-rich motif以外に、新たに負に働くシス因子を同定した。
