研究概要

1. β型トランスフォーミング増殖因子(TGF-β)のII型レセプターによるI型レセプターの活性化
TGF-βはまずII型レセプターに結合し、この結果I型レセプターをリン酸化して活性化するが、そのさいI型レセプターのGSドメインとよばれる構造が重要であることがこれまでに知られている。今回我々は、I型レセプターの膜貫通領域とGSドメインの間に位置する領域(以下、JM領域)のシグナル伝達における重要性を明らかにした。さらにII型レセプターとI型レセプターのリン酸化部位の同定を試み、II型レセプターでは膜貫通領域とキナーゼ活性領域の間に位置する領域とC末端領域が、またI型レセプターではJM領域とGS領域のセリンやスレオニンがリン酸化されることを確認した。今後はリン酸化によってどのようなシグナルが伝えられるかを明らかにする。
2. TGF-βI型レセプターの細胞内基質の同定とシグナル伝達機構
我々は酵母を用いたtwo-hybrid法によってTGF-βI型レセプターの基質となるペプチドのクローニングを試みた。その結果FKBP12(免疫抑制剤であるFK506やラパマイシンの細胞内結合ペプチド)やfarnesyl transferase-αを見出した。さらに最近ショウジョウバエ由来のI型レセプターであるthick veinsやsaxophoneに結合するタンパク質の同定をtwo-hybrid法で試みた。その結果、FKBP-12の他、少なくとも3つの新しいタンパク質が結合することが明らかとなった。今後はこれらのタンパク質のシグナル伝達における役割、とくに増殖抑制やアポトーシスへの効果を検索する。
