低分子量G蛋白質Rasを介するシグナル伝達機構とその病態

研究概要	低分子量G蛋白質Rasは複数の標的蛋白質を介して、種々の細胞機能を制御することが明らかになりつつある。本研究ではRasのシグナル伝達機構の全貌を明らかにすることを目的としている。私共はこれまでにRasの標的蛋白質であるRafが14-3-3蛋白質と結合することを明らかにしている。本研究において、COS細胞内でRasはRaf(1-135)(Rafのアミノ酸1から135番)と、14-3-3蛋白質はRaf(136-322)と結合し、Sf9細胞内でRafはRasと14-3-3蛋白質に同時に結合した。これらの結果から、RafのRas結合部位と14-3-3蛋白質結合部位は異なっており、Rasと14-3-3蛋白質の両蛋白質がRafに同時に結合することによりRafが活性化される可能性が示唆された。私共はRalGDSがRasの新しい標的蛋白質であることを提唱しているが、本研究において、c-RasとRalGDSをトランスフェクションしたCOS細胞をEGFで刺激すると、EGFの濃度依存性にRalGDSはc-Rasと結合した。AキナーゼはRafを直接リン酸化して、Rasとの結合性能を抑制し、その結果RasからRafへのシグナル伝達系を阻害することが知られている。RalGDSもRafと同様にAキナーゼによりリン酸化されたが、RalGDSのリン酸化はRalGDSのRas結合活性に影響しなかった。さらに、Aキナーゼの活性はEGF依存性のRafの活性を抑制したが、RasとRalGDSの結合には影響しなかった。以上の結果から、Rasの細胞内シグナル伝達機構はRafを介する経路とRalGDSを介する経路が存在することが決定的になった。また、RasからRafとRalGDSに伝達されるシグナルはAキナーゼにより選択的に制御されていると考えられる。私共はRalGDSのC末端側にRasと結合する部位(RID)が存在することをすでに明らかにしている。活性型RasでトランスフォームしたNIH3T3細胞にRIDを導入すると、低濃度血清中での増殖能や寒天培地中での増殖能が抑制された。しかし、活性型RafでトランスフォームしたNIH3T3細胞にRIDを導入しても、変化は認められなかった。これらの結果はRalGDSが細胞内でRIDを介してRasと結合し、そのシグナルを伝達することを示すものである。さらに、RIDはRasの機能阻害剤として、シグナル伝達機構の解析やがん治療薬の開発へ応用できる可能性があると考えられる。以上、本年度の研究は予定通り進行していると判断しているが、来年度もさらに詳細な検討を続けて、Rasを介するシグナル伝達機構を明らかにし、その異常による病態を解析したいと考えている。
