赤痢菌の上皮細胞侵入初期過程の分子機構

研究概要	赤痢菌の細胞侵入初期過程の分子メカニズムを理解する目的で、本菌の分泌するIpaB,IpaC,IpaD蛋白の細胞侵入に於ける菌体表層上の動態、およびエンドサイトーシス誘発に至るIpa蛋白と上皮細胞表層の相互作用を解析した。具体的には、Ipa蛋白の菌体からの遊離を司るSpa因子の機能解析と、上皮細胞側のIpa蛋白の菌体遊離促進化因子の探索、及び遊離Ipa蛋白に対する宿主細胞レセプターの同定を実施し以下の知見を得た。(1)Ipaの菌体からの遊離にはSpa蛋白の一つ、Spa32が関与していた。(2)Spa32は分子内にジスルフィド結合を一つ有し、その湾曲構造はSpa32の外膜発現およびIpaの細胞表層から外液への遊離に必須であった。(3)Ipaの遊離は赤痢菌と上皮細胞の接触により著しく促進された。(4)極性化した上皮細胞へ赤痢菌を接触させると、頂端側に比べ基底膜側からの細胞侵入能は数百倍高く、遊離Ipa蛋白量もそれに比例した。(5)赤痢菌を細胞外マトリックス(フィブロネクチン、ラミニン、コラーゲンタイプIV)に接触させると、Ipa蛋白が遊離した。(6)遊離Ipa蛋白は、IpaB,IpaC,IpaDからなる複合体を形成した。(7)Ipa複合体は人α5β1インテグリンと特異的結合性を示した。(8)Ipaとインテグリンの結合は、赤痢菌の細胞侵入を促進した。(9)Ipaとα5β1インテグリンの結合は、水溶性フィブロネクチンの添加では阻害されず、組織型フィブロネクチンにより著しく阻害された。以上の知見は、赤痢菌の上皮細胞への侵入はSpa32により菌から遊離したIpa蛋白複合体と宿主細胞のα5β1インテグリンの結合が重要であることを強く示唆している。
