研究概要

NSP1遺伝子に500塩基の欠失あるいはORFの途中にナンセンス変異をもつウシロタウイルス株A5-10、A5-16の解析の結果、以下の知見を得た。
1.A5-10またはA5-16感染細胞において、NSP1蛋白(低分子量断片も含め)の発現がなく、これまで多くの研究者より強調されていたロタウイルスの増殖におけるNSP1蛋白の役割について再考する必要性を提起した。
2.A5-10またはA5-16のNSP1 cDNAを用いたin vitro translationでもNSP1蛋白の発現がなく、1.の結果を支持した。
3.NSP1遺伝子の欠陥にもかかわらず、A5-10、A5-16は乳のみマウスに下痢を起こした。こうして、ある条件下ではNSP1の機能がロタウイルス増殖過程で発揮される必要性がないのではないかと考えられた。
4.SA11-S1株とA5-10、A5-13、A5-16との混合感染の結果得られたリアソ-タントの解析の結果、A5-10、A5-16のNSP1遺伝子は、その選択が著しく低く、NSP1遺伝子の機能は他セグメントのバックグランドに大きく左右されることが判明した。
現在、A5-10、A5-13、A5-16のNSP1遺伝子のバキュロウイルス系での発現に向けた準備(トランスファーベクターへの導入その他)が進行している。また、A5-10、A5-16のNSP1蛋白に対応する合成ペプチドに対する抗血清は、NSP1の抗原性が弱く満足すべき血清が得られていないが、領域を検討し再度試みる予定である。
