TypeIII受容体チロシンキナーゼの造血・免疫組織発生における役割

研究概要

1、昨年度に作成したFlk1を認識する抗体を用いて、血管・造血系細胞が中胚葉より発生してくる過程を追跡するための実験系を確立すべく実験を行った。この結果、ES細胞からproximal lateral中胚葉をほぼ選択的に誘導する培養系、その間の細胞表面マーカーの変化、そしてFlk1,E-Cadherinに対する抗体を用いた、proximal lateral中胚葉の純化、最後に、純化された中胚葉細胞から血管や血液を誘導する培養方法について全て確立することができた。今後、このプロセスを調節している新規分子の同定を順次行っていく予定である。
この方面の進展と比べると、Flk1分子の機能を阻害するモノクローナル抗体の確立は進展していない。
2、IL-7Rに対する抗体を用いて昨年開始したパイエル板の発生過程についての研究は大きな進展を見た。即ち、マウスパイエル板形成が胎生14-15日よりまずVCAM-1,続いてIL-7R,CD4,Ia陽性細胞の局所への集積、そして最終的に成熟リンパ球の血管を介する移動の3段階を経て起こることを明らかにした。さらに、このVCAM-1の発現で代表されるパイエル板原基の形成と維持にIL-7Rを発現する未熟リンパ球が必須であることを明らかにできた。このシナリオに基づいて、IL-7Rに対する抗体を胎児に投与する実験を行い、パイエル板が欠如している一方、他のリンパ組織はまったく正常の実験動物を創出できることを明らかにした。このモデルマウスは、今後の腸管免疫機能の解析に特に重要であろうと予想している。
3、胎児肝での、adult typeの赤血球発生に、PDGFRa陽性ストローマ細胞の形成が必須であることを証明した。
4、c-fmsの機能抑制実験より、成熟後の骨代謝バランスの維持にM-CSFが必須であり、このバランスの異常により、人でosteopoikilosisとして知られる病態が誘導されることを明らかにした。
