研究概要	B細胞抗原受容体のシグナル伝達とその活性化のメカニズムを明らかにするため、B細胞レセプターシグナル伝達の新規な機能分子の解析を行った。抗原レセプターコンプレックスのIgα(MB-1)に結合するリン酸化蛋白p52をみつけ、この分子をコードするα4遺伝子の構造、発現、機能の解析を進めた。この分子は未熟なステージのB前駆細胞、pre-B細胞、sIgM^+未熟B細胞の時期からすでに発現しており、これらの細胞で未知のリン酸化酵群と強固に結合しているシグナル伝達中継分子である可能性が示唆された。正常休止期のB細胞では通常結合リン酸化活性はほとんど検出できないが、抗原レセプターシグナルによって著しく上昇することを見つけた。また、この酵素活性は細胞周期の回転と密接に関連していることから、このα4遺伝子を介するシグナルがB細胞の増殖に重要な機能をする経路であることが考えられた。さらにα4遺伝子の酵母菌におけるホモローグTAP42はラパマイシン感受性経路で、細胞の栄養刺激から誘導される増殖反応に関与することから、リンパ細胞におけるラパマイシン経路との関連性について詳細な検討をおこなった。その結果、α4遺伝子及びコードする蛋白はリンパ細胞のラパマイシン感受性の経路に関連することが明らかになった。類似した免疫抑制剤サイクロスポリン A,FK506のシグナル伝達経路ではカルシニューリン(phosphatase2B)との結合を阻害することでT細胞シグナル伝達をブロックするが、α4分子はこの経路とは異なるphsophatase2Aを介する経路に関与していることが示唆された。この経路の解析を進め、免疫抑制剤の作用点を同定することが可能となると考えている。
