研究概要

B細胞活性化シグナルの解析を行い、リンパ細胞増殖関連シグナル伝達分子α4の固定、遺伝子解析、およびα4分子のリンパ細胞シグナル伝達分子における役割を解析した。
マウスのα4、ヒトのα4分子のcDNA解析、染色体遺伝子構造の解析、遺伝子発現調節の解析を行った。ヒトではX染色体に位置し、マウス同様リンパ細胞の増殖に関連する分子であることが示された。
α4分子の機能を解析し、この分子が細胞内でタンパク質脱リン酸化酵素PP2Aの触媒ユニットと直接結合する分子であることを示した。その結合ドメイン、リン酸化部位、コンセンサス領域を検討した。免疫抑制剤ラパマイシンに感受性経路のシグナル伝達分子であることが明らかになり、α4分子とPP2A分子との直接の結合が、ラパマイシンによって制御されることも示した。α4/PP2A分子はラパマイシン結合分子からのTor(Target of Rapamycin)の下流に位置するリンパ細胞増殖制御シグナルを伝達する機能分子であることが、示された。
免疫制御剤ラパマイシンのリンパ細胞における作用点を明確にしたとともに、新たなリンパ細胞増殖シグナル伝達経路を明示した。現在、この分子の果たす免疫応答における役割について解析を進めている。
