肺サーファクタント蛋白質Aおよびその受容体遺伝子を用いた肺病態分子機構の解析

研究概要

肺サーファクタント・アポ蛋白質は、肺胞細胞におけるリン脂質代謝を調節するとともに肺における生体防御機構にも大きく寄与している。本研究は、C末端側にC型レクチン構造を有する「コレクチン・ファミリー」に属するサーファクタント・アポ蛋白質SP-Aとその受容体に着目し、肺胞細胞の機能調節におけるSP-A関与の分子機構を解析することを目的としている。平成7年度の研究実績を以下に要約し、本研究で得られた新知見を列記する。
1.SP-Aとともにコレクチン・ファミリーに属するマンノース結合蛋白質(MBP)は、SP-Aと異なり、ホスファチジルコリン結合能や肺胞II型細胞との相互作用を有していない。そこで、SP-AのC末端側をMBPと置換したSP-A/MBPキメラ分子による検討で、SP-Aの脂質結合部位、および、肺胞II型細胞との相互作用に直接関与する部位は、SP-A分子のGlu^<19 5>からC末端側(Phe^<22 8>)には存在せず、ネック領域からC末端側のGly^<11 5>〜Gly^<19 4>の領域が重要であることが示唆された。また、正電荷を持ったLys^<20 1>とLys^<20 3>をそれぞれAlaに変換させた変異体はともに生物活性を有していたので、Lys^<20 1>とLys^<20 3>はSP-Aの機能発現に関与しないことが判明した。
2.肺胞細胞より形質膜を分離し、種々の界面活性剤を用いて膜の可溶化を検討したところ、オクチルβD-グルコピラノシドとトリトンX-100が肺胞細胞の膜蛋白質の可溶化に適していることが判明した。
3.肺胞蛋白症患者の気管支肺洗浄液から分離した病的SP-Aには2種類の分子サイズが存在し、Rotary Shadow Techniqueを用いたSP-A分子の電子顕微鏡観察で、正常SP-Aで観察される18量体のほかに、18量体が2〜3個集合した多量体SP-Aが同定された。正常肺胞腔に観察される格子様の管状ミエリン構造はSP-A、SP-Bとリン脂質で再構成できるが、病的な多量体SP-Aは管状ミエリン構造を形成できなかった。また、肺胞II型細胞との相互作用においても、この病的多量体SP-Aは、サーファクタント・リン脂質分泌抑制能、受容体結合能ともに、18量体を示す正常SP-Aに比べて、著しくその機能が劣っていた。
