遺伝子ターゲティングによる血管壁コレステロール蓄積機構の解明

研究概要	粥状動脈硬化症の形成には大きく二つの現象が関与している。一つは血管壁におけるコレステロールエステルの蓄積であり、一つは血管壁における細胞成分の病的反応である。どちらが欠けても粥状動脈硬化症は成立しがたい。特に脂質代謝異常は最大の危険因子と考えられており、脂質の蓄積は、動脈硬化症の初期病変である泡沫細胞の形成に関与している。私共は細胞内コレステロール代謝調節という観点より、新しい粥状動脈硬化症の内科的治療法の開発に発展したい。そこで、細胞内コレステロール代謝調節に関与するACAT及びNCEHのgene targettingを行うことにより、血管壁コレステロール蓄積機構を明らかにしようとしている。またホルモン感受性リパーゼのgene targettingを行うことにより、本酵素とNCEHの同一性、即ち脂肪細胞での脂肪分解と血管壁でのコレステロールエステル分解反応のクロスオーバーについて、遺伝子レベルで明確にしようとしている。既にACAT及びホルモン感受性リパーゼ(HSL)のcDNAクローニングを終了し、gene constractを作製して、ES cellに導入し、マウスへの遺伝子導入に成功している。本研究を通じて、細胞内コレステロール代謝調節の異常や、随伴する血管壁における細胞成分の病的反応を明かにして、粥状動脈硬化症の成因を解明していきたい。特にHSLノックアウトマウスでは、肥満と動脈硬化症の進展にHSLがkey enzymeであることが示されつつある。
