研究概要	今年度は、血栓性素因protein S(PS)欠損症において、分子異常の同定ならびにPS欠損症に至る分子病態の解明を試みた。I型PS欠損症患者とその家族の凝血学的解析および患者PS遺伝子の解析を行い、Arg474(CGT)->Cys(TGT)の遺伝子異常を同定し本異常が家系内でのPS欠損症と一致して伝搬していた。この異常PS遺伝子は血小板においてmRNAレベルまで転写されており、Cys474-PSの発現実験より翻訳後の分泌異常と細胞内分解が欠損症の病態であると推察された。また、Arg474は関連蛋白でも種を越えArgかLysに保存されており、そのAla、Glu置換でも分泌異常が生じLys置換ではほぼ正常に分泌され、かつ非還元下SDS-PAGEでArg474-PSとCys474-PSの分子量に明かな差が認められたことより、この塩基性アミノ酸がPSの立体構造構築に重要な役割を果たしていることが示唆された。一方、血管内皮細胞ヘパラン硫酸プロテオグリカン・Ryudo canのヒト・ゲノム遺伝子をクローニングしてその性状解析を行なった。ヒトRyudo can遺伝子は5つのエクソンをもつ全長約23kbで、エクソンIは5′非翻訳領域とシグナルペプチド、エクソンII〜IVはグリコサミノグリカン結合部位をもつ細胞外領域、エクソンVはで細胞膜貫通領域、細胞内領域および3′非翻訳領域をコードしていた。プライマー伸長反応実験により第一ATGより43bp上流から転写が開始されていることが判明した。5′非翻訳領域は極めてG/Cに富み(72%)、転写開始点の25bp上流にTATA様配列(TATAAGA)が存在するほか、様々な転写因子の結合部位と推定される配列が存在した。また、Luciferase Reporter Vectorを用いた発現実験から5′非翻訳領域にプロモーター、エンハンサー、サイレンサーエレメントの存在が推測された。
