脂質性オ-タコイドによるネフロン機能調節の分子機構と生理的・病態生理的意義の解明

研究概要	脂質性オ-タコイドである血小板活性因子(PAF)やトロンボキサン(TX)A_2は腎での作用の分子機構や生理、病態的の意義について不明な点が多い。そこでラット、マウス腎のmicrodissectionによる各ネフロン分節、腎由来の培養細胞、受容体過剰発現及び欠損マウス及び糖尿病性腎症患者DNAを用いて、我々自身の開発した方法など近年の方法論的進歩を腎研究に適用した分子細胞生物学的、生理学的検討を行い、以下の知見を得た。1)PAF及びTXA_2受容体mRNAのネフロン各部位でMRT-PCRにより定量的な測定により広範なネフロン内での分布、特に尿細管での存在の証明及び近位尿細管でのPAF及びTXA_2アナログによる細胞内Ca^<2+>上昇の証明。2)培養メサンギウム細胞でのNorthern分析により、PAF受容体mRNAのPAF刺激により産生されるPGE_2によるhomologousなdown-regulationとそのcAMPを介する機構の証明。3)PAF受容体過剰発現及び欠損マウスにおける糖尿病性、シスプラチン腎症モデルの解析により、これら腎疾患モデルの病態でのPAFの関与の実証。4)PAFアセチルヒドラーゼの患者白血球DNAのPCR産物によるrestriction fragment length polymorphism解析による遺伝子多型、血清中の活性と糖尿病性腎症の腎疾患発症、進展の関連を検討した。結果はPAFの関与の傾向は認めるが、現在のところ統計的有意差は得ていないので検討継続中である。これらの結果は、脂質性オ-タコイドの新しい尿細管作用の発見、in situでの病態への関与更に関連した疾患治療の可能性を示したと言える。
