研究概要

1.発達期脳組織バンキングの進歩
胎児・新生児・小児の脳組織を診断・保存し、1000例を越した。これを用いて、神経病理学的、分子生化学的研究を行うことができた。
2.発達期脳の生化学的、分子生物学的ならびに免疫組織化学的研究の成果
細胞接着分子L1の発現について、ヒト脳の発生・発達の過程で観察した。L1は神経突起発生の初期に強く発現し、正常な神経細胞移動、神経突起の伸展および軸索の発達に関連し、正常指標となった。また、知能障害や難治性てんかんを呈する結節性硬化症の遺伝子蛋白tuberinの抗体を作製し、免疫化学的に検討し、腫瘍組織で低下し、腫瘍発生に関与していることを証明した。
脳性麻痺の原因となる脳室周囲白質軟化は脳低灌流によって起こり、サイトカインの一種であるTNFαは病巣部および周辺に増加し、ミクログリアを介して壊死の発生に関与していた。また、pontosubicular neuron necrosisは胎児・新生児に特有な神経細胞死であり、核崩壊像やグリアの反応の状態からアポトーシスに類似する形態を示した。この神経細胞死の初期にアポEが一時的に発現し、細胞死の防御に関与していると考えられた。新生児期の乳幼児突然死症候群は1ケ月以上の例と同じように、サブスタンスPと反応性アストロサイトの増加があり、出生前あるいは後の死亡前に慢性低酸素症があったことが考えられた。
