血流に起因する血管新生機序の生体工学的研究

研究概要	血管新生における血行動態の役割はいまだ充分に解明されていない。本研究では血流に起因するshearstressと血管新生の関係を明かにするため、慢性的な血流負荷が実験動物の血管構築に及ぼす影響を検討した。ウサギの耳介およびラット背部皮膚に微小循環観察用の透明窓(transparent chamber)を装着後、血流負荷群には血管拡張薬を持続的に投与して末梢血流を増加させた。生体顕微鏡ビデオシステムで経時的に窓内の微小循環を観察記録した。Dual window法を用いた血流測定システムで、記録されたビデオ画像から微小血管における赤血球速度を測定し、shearstressを算出した。また血管画像をデジタル化しコンピューター画像解析を使って血管構築および血管新生を定量した。パラメーターとして血管密度、総血管床面積などを求めた。血管拡張薬の持続投与によりshearstressの増大がみられた。負荷群において血管密度、総血管床面積のすべてが有意に増加した。ウサギの耳介透明窓では組織再生過程の血管新生が血流負荷により促進された。われわれは最近の内皮細胞に関する研究結果から血管系の最適分岐構造が内皮細胞のshearstress適応で実現されると考えている。本研究では皮膚・軟部組織の血管新生が血流負荷により促進されることが定量的に明かとなった。この結果は局所の血流に応じた血管構築を構成する制御機構の存在を示唆する。血管新生を生体の適応反応の一つとしてとらえることが可能と考えられる。すなわち負荷されたshearstressに対し血管新生が亢進することで応力が生理的最適値に調節されるという機構が実験的に示された。
