単クローン抗体を用いたヒト黄体細胞の分化抗原および分化機構の解析

研究概要	我々は、現在までほとんど解明されていなかった黄体の分化機構にさらに新しい糸口を見い出す目的で、ヒト黄体そのものを免疫原として黄体細胞に対する単クローン抗体作成し、顆粒膜・莱膜黄体細胞に特異的な細胞表面抗原をいくつか捉えることにした。ヒト黄体は性周期維持、及び妊娠初期の胎児維持に重要な役割を果たしており、その機能異常は、臨床的には黄体機能不全などの不妊症及び習慣性流産などの原因になると考えられている。また黄体は妊娠黄体への分化または黄体退縮等、急速な成熟・分化・細胞死をとげる特異的な内分泌細胞であり、細胞生物学的にも重要な細胞分化モデルとして注目されつつある。本研究は、黄体の分化に特異的な新しい抗原を単クローン抗体作成法で求めるもので、遺伝子を含めた抗原解析をめざし、現在まで情報の極めて限られていた黄体の分化関連物質に初めてメスをいれ、黄体の新しい分化機構を明らかにすることを目的としている。このような試みは世界初のものであり困難が予想されたが、現在、いくつかの抗体の作成に成功し、抗原を精製同定しつつある。そのうちのOG-1やHCL-1について得られた知見から、月経黄体から妊娠黄体への移行は単に内分泌的機能が維持されているだけではなく別の段階への分化であることを明らかにし、さらにこの分化を調節している因子は従来考えられていた胎児組織由来のゴナドトロピンだけではなく免疫学的因子が関与していることを報告した。また黄体形成期において細胞表面に発現している接着因子と細胞周囲に存在する細胞外基質との相互作用によって、黄体化が調節されていることを見いだしたが、これは局所における黄体化調節因子としては初めての報告となる。
