子宮内膜分化制御因子の解析による着床障害の病態解明

研究概要

まず、ヒト子宮内膜細胞の分離培養とプロゲステロンによる分化誘導増殖期を行った。分泌期初期のヒト子宮内膜より、間質細胞、腺上皮細胞を分離して培養した。プロゲステロン添加によって間質細胞を培養し、プロラクチン、IGFBP-1、relaxinなどを指標として分化の誘導を確認し、分化前後の培養細胞からRNAを抽出した。また、プロゲステロン無添加で同様に培養した細胞からもRNAを得、この両者よりcDNAライブラリーをそれぞれ作成し、次いで互いにサブトラクションを行って、分化によって増幅あるいは始めて出現する遺伝子群の分離濃縮を試みた。得られたcDNAライブラリー(subtracted cDNA)は、ベクターにグローニングして、Northern Blotによるスクリーニングした。約1,000種のクローン解析で、プロゲステロン依存性に出現する遺伝子が12種分離された。その中でも、培養初期に出現するクローンについて遺伝子解析を行った結果、tissue trans glurtaminase(TGase)とtissue inhibitor of metalloproteinase(TIMP)-3が同定され、その他の未知の遺伝子についてはさらに解析を進めている。前記のサブトラクション法で得られたTGase遺伝子は、内膜細胞の分化のごく初期より発現が急増し、一方TIMP-3はそれより遅れた培養6日後に発現していた(プロラクチンと類似した発現パターン)。機能を検討するために、TGaseの阻害剤であるmonodansilcadaverinを培養系に添加すると、プロゲステロンによる細胞の分化は添加濃度依存性に抑制され、またTGase mRNAのAntisense probe投与でも同じく分化抑制が見られた。これより、TGaseは分化過程の極初期より非常に重要な役割が想定される。そのため現在、vivoにおいてもこの酵素の役割を検討中である。
