骨再生過程におけるBMPの役割

研究概要

平成7・8年度の研究により以下の結果が得られた。
1)肋骨骨折部を形態学的に解析し、骨折後3日目に明らかな軟骨組織が出現することが明らかとなった。また、抜歯後4日目でalkaline phosphatase陽性の骨芽細胞様細胞が出現することも明らかとなった。
2)骨折1-2日目と4-5日目の肋骨骨折部よりそれぞれ細胞を採取し、培養し、骨芽細胞の性状を解析したところ、骨折4-5日目に採取した細胞の方がより強い骨芽細胞の性状を呈していることが明らかとなった。
3)骨折1-2日目ではBMP-2およびBMP-4のmRNAの発現がup-regulationされていることがRT-PCR法で確認できた。
4)ニワトリのSonic hedgehog(Shh)をtransfectionしたニワトリ線維芽細胞の培養上清はC3H10t1/2およびMC3T3-E1細胞のALP活性を促進した。
5)ニワトリのShhをtransdectionしたニワトリ線維芽細胞をヌードマウスの筋肉内に移植すると、骨形成が誘導された。
以上の結果から、本研究から次のことが示唆された。
1)骨折の治癒過程ではBMPが重要な役割を担っている。
