Wilms腫瘍の癌抑制遺伝子産物の転写制御活性

研究概要	これまでに、Wilms腫瘍のがん抑制遺伝子WT1の産物が転写因子で過剰発現すると細胞周期のG1期からS期への移行を抑制する活性をもつことを明らかにした。さらに急性白血病の症例ではWT1が発現しており、その発現レベルと予後が逆相関すること、急性白血病由来細胞株K5632の分化を誘導するとWT1の発現が低下し検出されなくなること、WT1のantisense oligonucleotideがK562細胞やAML患者由来細胞の増殖を抑制することが明らかになった。これらの結果から、白血病細胞ではWT1の発現が増殖にpositiveに作用している可能性があると考え、WT1を発現していないM1細胞にinducibleにWT1を発現させその増殖に及ぼす効果を検討した。その結果WT1を発現させるとM1細胞はG1 arrestとapoptosisを起こすことが見出された。したがって、WT1の作用は細胞の種類により異なり、単純に一般化はできないと考えられた。さらに他の細胞系を用いると、はじめに述べたようにNIH3T3細胞ではG1 arrestのみをおこし、F9細胞ではレチノイン酸で分化を誘導するときにのみapoptosisを起こすという異なった効果が観察された。いずれにしても、WT1はG1 arrestのみでなくapoptosisを起こす活性もあることが明らかになった。さらにWT1の機能を明らかにすることを目的としてWT1の作用によって発現の誘導される遺伝子をサブトラクション法による単離を開始した。
