ヘム酵素とその変異体ハイブリッドを用いた不安定反応中間体のキャラクタリゼーション

研究概要	ペルオキシダーゼやP450など酸化反応を触媒するヘム酵素は、活性発現のために高い酸化能力を有する高原子価オキソ-鉄錯体を生成していると考えられている。しかしながら多くの中間体は、未だ酵素系では観測されておらず、高原子価状態を生成する機構の詳細、特に活性部位周辺アミノ酸の役割解明が急務となっている。本研究では、モデル研究で示された反応機構に基づく活性部位周辺のアミノ酸の役割を実証する目的で、ミオグロビンの遠位ヒスチジンの空間的な位置を置き換えるミューテーションを行い、ミオグロビンに酸素酵素活性を賦与する研究を行っている。本年度は、ミオグロビンの64番目のヒスチジンをフェニルアラニン43の位置へ移動することで、酸素添加能が極めて高くなることを見いだし、本ミュータントを用いることで、反応活性種であるO=Fe(IV)ポルフィリンπラジカルの直接観測に初めて成功した。本活性種は60ミリ秒程度で生成し、数秒以内に一電子還元を受けてO=Fe(IV)ポルフィリンへと変化する。ダブルミキシング方によって種々の基質と反応活性種との反応を直接観測することにより、詳細な反応性の検討を現在行っている。一方、不安定化学種を低温下、有機溶媒中で直接観測する目的で西洋ワサビペルオキシダーゼの蛋白表面の化学修飾を行い、有機溶媒中で高原子価状態にある中間体の初めての観測に成功した。
