多能性造血幹細胞の分化系列決定の間質細胞による制御

研究概要

本研究では,樹立した2種の間質細胞株をフィーダーとして、セルソーターにより純化した骨髄造血幹細胞を培養し、新たなサイトカイン等の添加なしに起きる幹細胞の増殖と、ミエロイド系列分化誘導とBリンパ球系列分化誘導を規定する間質細胞の因子を同定し、造血幹細胞分化系列決定の制御機構を解明することを目的として、セルソーターを用いて純化した造血幹細胞を2種の骨髄間質細胞株と共培養し、以下の結果を得た。
(1)TBR59株上では、幹細胞はミエロイド系列とリンパ球系列両方向に分化誘導され、間質細胞との直接の接着を介したシグナルが重要であることが分かった。また、オンコジーントランジェニックマウス由来の幹細胞よりこの細胞依存に増殖し、造血幹細胞の表現型を維持している不死化した細胞株が樹立できた。
(2)TBR31-1株上では幹細胞はBリンパ球のみへと分化決定が起きること、また、この培養系から、間質細胞株依存に増殖し続けるBリンパ球系細胞であることが分かった。また、この細胞をマウスに移植しても増殖・分化が認められた。
(3)TBR59,TBR31-1株に特異的なモノクロナール抗体をスクリーニングし、いくつかの特異抗体が得られた。また、それぞれのcDNAライブラリーを作製し、特異抗体が認識する分子の遺伝子クローニングが可能となった。
