短時間前脳虚血に伴う海馬CAl錐体細胞のアポトーシスとその回避

研究概要	1)nedd-2類似遺伝子のクローニングと虚血モデルにおける発現:私達は、短時間前脳虚血に伴うCAl錐体細胞の細胞死がアポトーシスであること(J Neuroscience,1995)を明らかにしてきたが、今回、同細胞死で発現してくる遺伝子を検討する目的で、マウスnedd-2類似遺伝子のラットのクローンを取り、その発現を検討した。ラットのnedd-2類似遺伝子は構造的にはマウスと同じであった。砂ネズミの虚血モデルで検討したが、陽性像は得られなかった。2)細胞死回避因子の同定:私達はPCl2細胞に細胞死を回避する遺伝子として知られるbcl-2を組み込み(PC-bcl)、細胞死の回避実験を行なった。PC-bcl細胞は無血清下で生存すると共に、神経細胞様の突起を伸展した(J Neurobiol,1994)。この培養上清で無処置PCl2細胞を培養すると、無血清でも生き続け突起を伸展する。この神経栄養因子(NGFではない)を同定するため、野性型PCl2細胞を用いたアッセイ系を確立した。野性型PCl2細胞を無血清条件下で培養すると(24穴プレート、5000 cells per well)、24時間内にほぼ細胞は死に至る。これに、PC-bcl細胞の培養上清を添加すると、その量に依存して細胞死が回避されることが分かった。細胞死回避の判定は、生存細胞の数を計測するのと、生存細胞のLDH活性を測定する方法で算定した。100倍に濃縮した培養上清を逆層カラムで分画すると分子量10kD以下、30kD、70kDの近傍に細胞死を回避する活性があることが分かった。3)消化管上皮細胞の細胞更新に伴う細胞死の検討:短時間虚血に伴う海馬CAl錐体細胞のアポトーシスに際して、死に行く細胞のリソゾーム系が活性化され、細胞は萎縮する。ヒト小腸絨毛先端部の上皮細胞は細胞死に陥り管腔に脱落する。この過程で、死に行く細胞は基底側の細胞膜を細胞質に取り込み(inward blebbing)、上皮細胞と基底膜との接触を断つ。これに伴い、先端部の細胞は、CAl錐体細胞と同様に、リソゾーム系が発達し、上皮細胞は萎縮する。核の形態変化は脱落直前に起こる(Arch Histol Cytol,1995)。
