研究概要

シナプトタグミンはシナプス小胞膜上に存在し、神経伝達物質放出調節に関与する蛋白質である。我々は、このシナプトタグミンにイノシトールポリリン酸が結合し、これによって、神経伝達物質放出が抑制されることを見出し、イノシトールポリリン酸/シナプトタグミンの神経伝達物質放出機構における新しい可能性を報告してきた。
我々は、このシナプトタグミンを介したイノシトールポリリン酸による神経伝達の阻害機構を分子レベルで明らかにする目的で、シナプトタグミンに結合し、イノシトールポリリン酸でその結合が阻害される蛋白質の存在を想定し、その精製を試みた。
マウスシナプトタグミンIIのイノシトールポリリン酸結合領域を含む部分をGSTとの融合蛋白質(GST-SytIIC2AB)として発現、精製し、この融合蛋白質に特異的に結合する蛋白質(SBPs)をマウス全脳の粗抽出液中から、glutathion SepharoseでGST-SytIIC2ABと共沈させた。このSBPs/GST-SytIIC2AB/glutathion Sepharoseから100μMIP6で溶出した画分中には複数種の蛋白質が存在し、その中にはclathrin assembly complex(AP2)の他、66kDa,80kDaのそれぞれ新規な蛋白質が含まれていることがSDS-PAGE,Westem blotting,部分アミノ酸配列解析から判明した。
