研究概要	エレクトロポレーション法をキイロショウジョウバエへの遺伝子導入に利用できるようにするための予備的な実験でアミラーゼ遺伝子を導入した結果、幼虫でアミラーゼ活性が検出されていた。本年度はショウジョウバエ受精卵をP-elementベクターとヘルパーのDNA溶液を含む緩衝液中に懸濁し、矩形波をかけてその後発生した幼虫に遺伝子導入がなされたか調べた。その結果、数千個体のオーダーでは形質転換個体を得ることができなかった。現在、様々な対照実験を行っており、原因を探っている。蚊科へのトランスポゾンを用いた形質転換の実験における宿主として、実験上様々な利点(飼育、卵の産卵形態、保存性)を持つヒトスジシマカ(Aedes albopictus)を用いた。生理食塩水を卵に注入した予備実験でふ化率13/231(5.6%)と効率はあまり良いとは言えないが、注入可能であることを確かめた。ショウジョウバエで広く行われているように眼色のマーカー遺伝子をベクターに用いるため、EMSを使ってヒトスジシマカでの変異体の作成を行った。その結果褐色眼である、brown eye 1,brown eye 2の2系統の突然変異体が得られた。brown eye 1は性染色体である第一染色体、brown eye 2は性染色体以外のだ第二または第三染色体にあることが交配によりわかった。カイコのトランスポゾンに関する研究では、カイコのSINE様因子であるBm 1の完全型の起源を知り、この因子の転移を支配する逆転写酵素の由来を明らかにするために、Bm1.1を含む31個の独立なクローンを単離し塩基配列から、近隣接合分子系統樹を作成したところ、3種類のクラスターが出現した。これは、Bm1.1が少なくとも3種の原形を出発点として、逆転写過程を経てゲノム中に拡散したことを強く示唆している。
