固相でのmRNA高感度検出法の開発とマウス膵臓に対するプロラクチン作用の解析

研究概要	本研究はまず生体内に微量しか存在しない生理活性物質の検出法を開発し、それを用いマウス膵臓にあって重要な生理作用を営んでいると考えられるプロラクチン受容体タンパク質、あるいは受容体遺伝子の発現様式を明らかにすることを目的とした。微量物質の検出法や定量的解析法の確立を目指す第一歩として、プロラクチン受容体cDANおよびcRNAを競合試薬として用いた液相競合RT-PCR法によるプロラクチン受容体mRNAの定量法を確立した。この方法を用い単離培養膵島におけるプロラクチン受容体mRNAの発現に対する種々のホルモンの影響を調べたところ、プロラクチン受容体mRNAはプロラクチン自体によって正の発現制御を受けていることが明らかとなった。またスプライシングによって生じたプロラクチン受容体のサブタイプのmRNAの量比を定量する簡便法「Onesided competitive PCR法」を開発し、培養膵島におけるプロラクチン受容体mRNAの発現様式の研究に適用した。その結果、膵島では肝臓と異なり長型とよばれる機能型のmRNAが多く、プロラクチンの作用によって正の発現調節を受けていることが明らかとなった。このことから、膵島ではプロラクチンに対する細胞の感受性が、プロラクチン自体によって制御されることが示唆され、これらの成果を論文として公表した。次いで、組織切片およびメンブレン上でのmRNAとタンパク質の高感度増幅法を確立し、マウス膵臓組織における各プロラクチン受容体の分布および性状を知ることを試みた。我々は予めメンブレン上に固定したプライマーを用いる方法を着想し、すでに固相でのPCR法によるゲノムDNA断片の検出することは可能にしたが、mRNAの検出を成功させるには至っていない。現在はmRNA・cDNAを保持しつつ、逆転反応の効率を向上させるような反応条件を模索している段階である。
