脂肪交雑肉牛の肥育前検定法の確立

研究概要	和牛肉の品質評価では「霜降り」と呼ばれる筋肉と脂肪がきめ細かに交雑した肉質が珍重されている。この脂肪交雑肉牛生産の資金的リスクを回避するために、肥育前の素牛における肉質検定法を検討して以下の実績を挙げた。肉牛生産における集約的肥育過程では、エネルギー源の過剰投与で多分化能を持つ間葉系幹細胞の脂肪細胞への分化が筋肉組織内で促進されると考えられる。この脂肪細胞分化は多段階の決定を経て達成され、脂肪細胞の分布パターンは肥育前の素牛で決定されると考えられる。そこで、培養条件下で脂肪細胞に分化するマウス由来の3T3-L1及び3T3-F442A細胞株による基礎実験を行い、この細胞を皮下移植すると、マウス体内の所望の部位に脂肪組織を誘導できることを明らかにした。さらに、各種の成長因子を基底膜成分と混合して皮下注入すると、内在性の間葉系幹細胞の遊走・増殖・分化を誘導できることも発見した。注入する成長因子の種類によっては筋肉組織も誘導でき、脂肪交雑肉を人工的に形成させる組織工学の方途を開いた。また、脂肪交雑肉牛の肥育前検定の指標とすべく、脂肪分化の早期に発現する遺伝子転写制御因子であるPPARγ2,C/EBPα,C/EBPβ及びC/EBPγのウシ遺伝子群の単離にも成功した。これめらの組み換え体を大腸菌体内で発現させて、特異抗体を作成することにも成功した。これらの抗体を用いた免疫組織化学によって、脂肪交雑肉牛を予備的に検定する方法をほぼ確立した。
