研究概要	イネでは、アグロバクテリウムによる遺伝子導入は通常は成功しないとされているので、イネ細胞のプロトプラストに電気的に遺伝子導入する方法(エレクトロポーレーション法)によって形質転換を行い、その細胞を培養して一旦カルス様の組織をつくり、そこから個体を再生した。このようにして再生できた個体は26あった。それらの個体を培地上で育てた後通常の土壌栽培に移し、栽培したところ、実際に種子をつけたのはそのうち6個体であった。得られた種子を通常の土壌によるポット栽培したところ、そのうち1株が著しく窒素吸収が高いことを示す結果を得た。すなわち、分けつ期の頃まではほとんど対照区と差がなく成長したが、その後徐々に形質転換体では緑色が強くなり、分けつ数も20本と、通常の6-7本の約3倍となり、その後正常に出穂した。1穂当たりの籾数も平均して1-2割多くなった。図3を見ても分かるとおり、対照区のイネの茎の部分が黄色となっているのに、形質転換体では、緑色がきわめて強いことが分かる。これは、養分吸収の差を見るために、播種後肥料を全く与えなかった条件下での観察結果である。詳細な窒素吸収のデータは、個体数が多く準備されれば得ることができる。成長の観察という非常にシンプルな方法ではあるが、今回得られた結果は、当初の目標どおり、バイオテクノロジーの手法で窒素吸収・同化の鍵酵素量を増やすことにより、植物の窒素吸収・利用の能力を高めることができることを示すことができた。
