エンドトキシンショックの病態生理の解明と治療への応用

研究概要	エンドトキシンショックは、主としてグラム陰性菌の感染により、その外毒素による循環不全や全身での血液凝固を主症状とする致死的病態である。この原因としては、細菌膜に存在するリポ多糖類(LPS)が何らかの機序で、サイトカインやTNFを放出するためと考えられているが、詳細は全く不明である。エンドトキシンショック発生機序の解明に向けて、以下の成果を得た。我々はPAF受容体を単離し、エンドトキシンがこの受容体と結合することを明らかにし、また、エンドトキシン固有の受容体の存在を示唆する証拠を得ている。 (1)エンドトキシンショックモデル動物の作製 PAF受容体を高発現したマウスを作製した。このマウスは少量のエンドトキシンで死亡するなど、モデル動物として適当と考えられた。現在、死因を組織学、生化学等の手法で解析している。さらに、種々の薬剤による予防効果についても検討している。一方、PAF受容体を欠損したマウスの作製に向け、準備が進んでいる。 (2)エンドトキシン固有の受容体の単離エンドトキシンはMAPキナーゼを活性化し、この反応は種々のキナーゼが関与している。そこで、MAPキナーゼの簡易で高感度の測定法を開発し、これによりマクロファージ由来のcDNAをトランスフェクトした細胞でのキナーゼ活性化を指標とした発現クローニングが可能となった。以上のようにエンドトキシンショックの発生機序の解明と治療法の開発に関する準備が整った。
