キャピラリー電気泳動法を用いた遺伝子診断の自動化の開発

研究概要	高分子ポリマーを含む緩衝液により生じる分子ふるい効果を利用したNon-Gel Sievingキャピラリー電気泳動法を用いた遺伝子解析法を確立し、大腸癌臨床検体より抽出したDNAによる遺伝子診断に応用した。具体的には、遺伝性大腸癌および散発性大腸癌において重要な遺伝子マーカーであるp53遺伝子やTransforming Growth Factor-β Type II Receptor(TGF-β RII)遺伝子を対象としてその変異を検出する方法を確立した。p53については、泳動中のキャピラリー管内の温度を冷媒を用いて制御することによりSingle-Strand Conformation Polymorphism(SSCP)による点突然変異の解析を30分程度で実施できる系を確立した。更に解析したDNAを直接キャピラリーより分取しサイクルシークエンシング法にてその変異した塩基配列を決定できる系を開発し、臨床検体の分析に利用した。また、溶液内でハイブリダイゼーションを行い、形成されたハイブリッドDNAを検出できる条件も確立した。これは、変異や欠失など様々な遺伝子上の変化の解析に有効と考えられた。一方、TGF-β RII遺伝子を対象として、大腸癌などで配列の異常が検出される。この遺伝子上の繰り返し配列(A)repeatsにおける1塩基の欠失を分析できる系を確立した。この方法では、まず適当な条件下で非対象PCRを行い、(A)repeatsを含む配列を1本鎖DNAとして増幅させた。尿素およびCellulose系ポリマーを含む0.267Mトリス/ホウ酸緩衝液を用いたキャピラリー電気泳動法により1塩基の違いを分離できる系を確立した。この系を用い増幅した1本鎖DNAを解析し、(A)repeatsにおける1-2塩基の欠失を20-30分程度で検出できた。更に、遺伝性大腸癌を含む検体で異常を検出することができた。また,これらの方法では連続的に検体を分析できた。また、何れの方法においても再現性は、変動係数1.5%以内であり再現性も良好であることが確認された。このように、キャピラリー電気泳動法を用いた臨床検体を解析できる系を開発し、臨床応用可能な状況にした。
