研究概要	高親和性GM-CSFレセプター(GMR)はα鎖β鎖(βc)の二本鎖から構成され、β鎖はIL-3、IL-5と共有される。 GM-CSFの刺激に伴いJAK2キナーゼが活性化される事が報告されているが、各種のシグナル伝達反応での関与は不明であった。そこでJAK2のキナーゼドメインを欠失した変異体(ΔJAK2)を作製しBA/F3細胞に一過性に発現させ、GM-CSFシグナルへの影響を観察した。COS細胞で過剰発現させた野性型JAK2は自己リン酸化される。さらにΔJAK2を共発現させることにより自己リン酸化はΔJAK2の用量依存的に減少しΔJAK2がdominant negativeに作用することが示唆された。BA/F3細胞に一過性に発現させたΔJAK2はGM-CSFによるチミジンの取り込み、c-fos、c-mycの活性化を阻害した。チロシンキナーゼ阻害剤であるgenisteinはGM-CSFによる細胞増殖とc-myc転写活性を阻害するが、c-fos、c-junの活性化には影響しない。JAK2のリン酸化にはGMRの細胞膜貫通領域の直下の部分(領域A)が関与するが、c-fos、c-junの活性化によりC末端側の領域(領域B)が必要である。従ってGMRβの膜貫通部分直下の領域で相互作用するJAK2キナーゼがGM-CSFの刺激により活性化されGMRβの細胞内領域をリン酸化しc-fos、c-junなどの活性化にいたることが予測された。一方細胞増殖とc-mycの活性化にはc-fos、c-junなどの活性化に必要とされる領域bは介さないことから異なるシグナル伝達経路の関与が結論された。
