研究概要	1.ターゲッティングベクターの作製: 129/Svマウス染色体DNAライブラリーより脳性ナトリウム利尿ペプチド(BNP)とC型ナトリウム利尿ペプチド(CNP)遺伝子をクローニングし、BNP遺伝子は第1及び第2エクソンを、CNP遺伝子は第1エクソンをネオマイシン耐性遺伝子にて置換したポジティブ・ネガティブ選択用のターゲッティングベクターを作製した。 2.ES細胞へのターゲッティングベクターの導入と相同組換え体の選別: ターゲッティングベクターをエレクトロポレーション法にてES細胞へ導入し、G418とGANCによるポジティブ・ネガティブ選択法とサザンブロット法にて相同組換え体を選別した。BNPは50クローンのうち7クローン、CNPは80クローンのうち1クローンの相同組換え体を得た。 3.BNPあるいはCNP遺伝子欠損マウスの作製: 得られたBNP及びCNPの相同組換え体を8細胞期胚にマイクロインジェクションし、生殖キメラマウスを作製した。このうちgermline transmissionが確認されたものは、CNPは1系統得られ、CNPは現在ヘテロ接合体とホモ接合体(遺伝子欠損マウス)の作製に成功している。BNPについても遺伝子欠損マウスの作製に向けて交配を継続している。 4.心房性ナトリウム利尿ペプチド(ANP)とBNP遺伝子の構造解析: マウスBNP遺伝子クローニングの過程でマウス第4染色体上にBNPとANP遺伝子が約12kbの距離に隣接して存在することが明らかとなった。更に、PCR法にてヒトBNPとANP遺伝子がヒト第1染色体上では約8kbの距離に隣接して位置することを証明した。以上より心臓ホルモンであるANPとANP遺伝子が同一染色体上に隣接して存在することが証明された。これらの成績は、ANPとBNPの遺伝子発現調節や遺伝子進化の解明に重要と考えられた。
