癌の新しい診断と治療のための磁性体ハイブリッドの開発

研究概要

1.微粒子磁性体の作製:安定でしかも加工しやすい磁性体として、ポリエチレングリコール磁性体を開発した。この磁性体は抗体との結合も容易で、その際に抗体の活性が失われることもなかった。また、抗体以外の蛋白質との結合も可能で、種々のタンパク性抗癌物質を結合することにより、新しい複合体の調整が可能となった。
2.ポリエチレングリコール化磁性体と大腸癌に対するモノクローナル抗体A7との複合体を作製した。この複合体はin vitroでA7認識抗原陽性癌細胞とのみ選択的に結合した。また、ヌードマウスにヒト大腸癌を移植し、磁性体-抗体複合体を投与してMRIにより局在を検討したところ、複合体が癌組織に選択的に集積していることが確認された。
3.ポリエチレングリコール化磁性体の抗原性は、無視し得るものであることが明らかになった。しかし、A7を結合した場合にはA7に対する抗体(Human Anti-Mouse Antibody)の産生が認められた。抗体もポリエチレングリコール修飾する事で、ある程度解決可能と思われた。
4.Neoprobe Deviceの導入により、投与した抗体が選択的に癌病巣に到達することが確認できた。投与からあまり短時間では検出が困難であることも明らかになり、磁性体-抗体複合体の検出のための貴重な基礎データが得られた。
