骨芽細胞と破骨細胞の機能発現調節機構の解析に基づく骨代謝評価系の開発

研究概要	骨芽細胞の分化決定機構を解析し以下の結果を得た。筋芽細胞(C2C12細胞)を骨誘導因子(BMP-2)で処理すると、筋細胞への分化は抑制され、骨芽細胞への分化が誘導された。C2C12細胞はBMPのI型レセプターBMPR-IAのmRNAを発現していたが、MBPR-IBのmRNAを発現していなかった。キナーゼ領域を欠失したBMPR-IAを大量に発現させた細胞では、BMP-2存在下でも骨芽細胞への分化は誘導されず筋細胞へ分化したが、この筋細胞へ分化はTGF-βにより抑制された。キナーゼ領域を欠失したBMPR-IAを発現させた細胞に、さらにMBPR-IAまたはTGF-βのI型レセプターTβR-1のcDNAを大量に発現させてBMP-2で処理すると、BMPR-IAを導入したときのみ骨芽細胞への分化が促された。以上の結果は、C2C12細胞におけるBMP-2による筋細胞への分化の抑制と骨芽細胞への分化誘導作用が、どちらもBMPR-IAを介したシグナルによって制御されていることを示唆する。破骨細胞の波状縁形成のメカニズムを解析し以下の結果を得た。破骨細胞は、インテグリンを介してRGD-配列を有する蛋白質を認識し、接着面に波状縁を形成した。波状縁のマーカーである液胞型H^+-ATPaseの局在より、破骨細胞の極性化は骨表面だけではなくプラスチックディッシュ上でも起こることが示された。この極性化に際し、細胞内シグナル伝達因子であるp130^<Cas>のチロシン残基が特異的にリン酸化された。コラーゲンゲル上や脱灰骨上では破骨細胞は波状縁を形成せず、破骨細胞は接着基質の物性(硬さ)を認識し極性化することが示された。また、純化した破骨細胞は単独では骨吸収能を発現できず、その骨吸収活性発現には骨芽細胞の共存を必要とした。骨芽細胞による破骨細胞の活性化には、破骨細胞との摂食が必要であった。
