定着因子融合タンパク質を用いた齲蝕の免疫学的予防法の開発

研究概要	本研究では、Streprococcus mutansの定着因子である菌体表層タンパク質抗原(PAc)の垂液結合領域(PAcA)と非水溶性グルカン合成酵素(GTF-I)のグルカン結合領域(GB)やスクロース結合領域(SB)を融合させたタンパク質(PAcA-GB及びPAcA-SB)を遺伝子工学的手法を用いて作製した。同融合タンパク質をウサギに経皮免疫して、抗PAcA-GB抗体と抗PAcA-SB抗体を調製した。これらの抗体のグルカン合成やS.mutans菌体付着に及ぼす影響をウサギ抗PAc抗体、抗PAcA抗体、抗GTF-I抗体などと比較したところ、以下の結果が得られた。 1)抗PAcA-GB抗体と抗GTF-I抗体はS.mutansの菌体遊離型と結合型のGTFによる非水溶性グルカン合成やGTF-Iによる全グルカン合成を強く阻害した。一方、抗PAcA-SB抗体は上記酵素標品によるグルカン合成にほとんど影響を及ぼさなかった。 2)抗PAcA-GB抗体、抗PAcA-SB抗体、抗PAc抗体及び抗PAcA抗体はスクロース非存在下におけるS.mutans菌体の唾液被覆ハイドロキシアパタイト粒子への付着を阻害した。一方、スクロース存在下におけるS.mutans菌体の唾液被覆ハイドロキシアパタイト粒子への付着は抗PAcA-GB抗体と抗GTF-I抗体によって強く阻害されが、抗PAcA-SB抗体、抗PAc抗体、抗PAcA抗体などによっては阻害されなかった。以上の結果から、抗PAcA-GB抗体はS.mutansの歯面への定着のコントロールに有用である可能性が示唆された。
