薬剤排出蛋白阻害剤スクリーニング系の開発

研究概要

7年度においては、まずTetとNorAを大腸菌において発現し、活性を解析する系の開発を目指した。なぜなら、当初の計画では、これらの排出系の性質を厳密に比較できるように、同一宿主での発現による解析を目指したからである。このうち、Tet Kについては大腸菌で構成的に良く発現する系が構築でき、輸送活性を詳細に解析することができた。この結果は報告した。NorAについても、大腸菌での発現系が構築でき、薬剤耐性をもたらすことが示された。ところが、反転膜を作成し輸送活性を反転膜での薬剤取込みとして測定することには成功しなかった。輸送活性がテトラサイクリン排出蛋白の場合よりはるかに低いことに主因があるものと考えられる。そこで、輸送活性については生菌体への薬剤蓄積量の低下を指標とすることにした。これには、排出蛋白発現株と発現しない感受性株に大きな差が認められる。ところで、反転膜を調整せず、生菌体で測定するのならば、スクリーニング系としては、臨床的に分離される状態に近いS.aureusでNorAおよびTet Kの活性を測定した方が良いと考えられる。そこで、これらの排出系の性質の基礎的な比較ならびに変異体構築などは大腸菌での発現系を用い、阻害剤のスクリーニングにはS.aureusでの発現系を用いることとした。緑膿菌のmexA/B系については引続きクローニングを試みるとともに、京都薬大の後藤博士よりプラスミドの提供を受けることとした。
